BACKGROUND: The study evaluated the effects of bacteriophage cocktail (BP) and ZnO administered during weaning time for piglets exposed to a non-sanitary environment. The bacteriophages were designed to eliminate Escherichia coli (K88, K99 and F41), Salmonella (typhimurium and enteritidis), and Clostridium perfreingens (types A and C). Forty 21-day-old crossbreed piglets were assigned to four treatments, including the PC (sanitary environment), NC (non-sanitary environment), BP (NC plus 108 pfu/kg BP), and ZO (NC plus 2,500 mg/kg ZnO). Piglets in the NC, BP and ZO were kept in a non-sanitary environment for 14 d, which was contaminated with the feces of infected pigs. RESULTS: Pigs in the BP and ZO treatments had a higher final body weight compared with the NC. The NC treatment showed the highest concentration of inflammatory cytokines including interleukin (IL)-1ß, IL-6 and tumor necrosis factor-α in the plasma. The administration of BP and ZO showed lower myeloperoxidase concentrations compared with the NC. The NC treatment showed a lower concentration of superoxide dismutase in serum compared with the PC. Among the treatments in non-sanitary environment, the NC treatment showed a higher concentration of malondialdehyde compared with the ZO. The PC treatment showed a lower concentration of butyric acid in the feces compared with the BP treatment. Among non-sanitary treatments, the villus height in the duodenum was greater in the BP and ZO compared with the NC. The lower abundance of Proteobacteria phylum was observed in the BP and PC treatments compared with the NC. The highest relative abundance of Eubacterium was recorded in the BP treatment. The abundance of Megasphaera and Schwartzia was higher in the NC pigs compared with the BP piglets. The abundance of Desulfovibrio was lower in the supplemented treatments (BP and ZO) compared with non-supplemented (NC and PC). The abundance of Cellulosilyticum genera was higher in the BP and ZO treatments rather than in the NC. The piglets in the NC treatment had the highest abundance of Escherichia-Shigella, followed by the PC and ZO treatments. CONCLUSION: In conclusion, these results suggest that the supplementation of bacteriophage cocktail could effectively control Proteobacteria phylum, Clostridium spp. and coliforms population and mitigated the adverse influences of weaning stress in piglets.

The purpose of this study was to compare marbling score, meat quality, juiciness, sarcomere length, and skeletal muscle satellite cell (SMSC) growth and related gene expression between Woori black pig (WB) and the Landrace, Yorkshire, and Duroc (LYD) crossbreed at different body weights (b.w.). WB was developed to improve meat quality and growth efficiency by crossbreeding Duroc with Korean native black pig. A total of 24 pigs were sacrificed when their b.w. reached about 50, 75, 100, and 120 kg. SMSC were isolated from the femoris muscles, and muscle and adipose tissues were sampled from the middle and the subcutaneous part of the femoris of hind legs, respectively. Expression levels of genes including Myoblast determination protein 1 (MyoD), Paired box gene 3 (Pax3), Myosin heavy chain (MyHC), and Myogenin, which are responsible for the growth and development of SMSC, were higher in LYD than the WB. Muscle growth inhibitor myostatin (MSTN), however, was expressed more in WB compared to LYD (p < 0.01). Numbers of SMSC extracted from femoris muscle of LYD at 50, 75, 100, and 120 kg b.w. were 8.5 ± 0.223, 8.6 ± 0.245, 7.2 ± 0.249, and 10.9 ± 0.795, and those from WB were 6.2 ± 0.32, 6.2 ± 0.374, 5.3 ± 0.423, and 17.1 ± 0.315, respectively. Expression of adipogenic genes in adipose tissue including CCAAT/enhancer-binding protein (CEBP)-ß, peroxisome proliferator activated receptor (PPAR)-γ, and fatty acid synthase (FASN), were greater in WB when compared with LYD (p < 0.01). Results from the current study suggest that different muscle cell numbers between 2 different breeds might be affected by related gene expression and this warrants further investigation on other growth factors regulating animal growth and development.

OBJECTIVE: For decades, LYD ([Landrace×Yorkshire] ♀×Duroc ♂) pigs are the most commonly-used commercial breed for meat production in Korea. Recently, due to the increasing demand for premium pork, the National Institute of Animal Science (Korea) has synthesized a novel pig breed named Woori Heukdon (WHD). This study aimed at comparing the carcass traits, meat yield and quality characteristics between the LYD and WHD pigs under identical rearing condition. METHODS: The WHD and LYD pigs (n = 15 each) were reared under identical conditions and fed the same commercial diet until reaching recommended market weight (100 to 120 kg). After slaughter, the carcasses were evaluated for traits and meat yield, and the meat quality was assessed on shoulder butt and belly cuts. RESULTS: Although no significant differences (p>0.05) occurred in slaughter weight between two pig types, WHD had a lower meat yield (by about 6 kg corresponding to approximately 7%) compared to the LYD pigs (p<0.05). The WHD had a higher fat content (by 4.26% and 13.52% in the shoulder butt and belly, respectively) compared to those of LYD pigs (p<0.05). The WHD meat showed a lower cooking loss and higher a* (redness) value (p<0.05). The WHD belly had a significantly (p<0.05) higher oleic acid content and concentrations of nonanal, octanal and decanal associated with fatty odor while, the LYD meat had a higher number of pyrazines associated with roasty odor. Regarding sensory quality, higher flavor, juiciness, and acceptability higher scores were given for the WHD meat than for the LYD meat (p<0.05). CONCLUSION: Under identical rearing conditions the WHD exhibited a better meat quality and sensory properties. However, the use of this diet resulted in a high fat level that may be associated with high trimming loss for the WHD.

Living systems provide a promising approach to chemical synthesis, having been optimized by evolution to convert renewable carbon sources, such as glucose, into an enormous range of small molecules. However, a large number of synthetic structures can still be difficult to obtain solely from cells, such as unsubstituted hydrocarbons. In this work, we demonstrate the use of a dual cellular-heterogeneous catalytic strategy to produce olefins from glucose using a selective hydrolase to generate an activated intermediate that is readily deoxygenated. Using a new family of iterative thiolase enzymes, we genetically engineered a microbial strain that produces 4.3 ± 0.4 g l-1 of fatty acid from glucose with 86% captured as 3-hydroxyoctanoic and 3-hydroxydecanoic acids. This 3-hydroxy substituent serves as a leaving group that enables heterogeneous tandem decarboxylation-dehydration routes to olefinic products on Lewis acidic catalysts without the additional redox input required for enzymatic or chemical deoxygenation of simple fatty acids.

This study was conducted to investigate the effect of cooling systems on reproductive performance, body temperature, blood metabolites, and the intestinal microbiome in early gestating sows exposed to high ambient temperature. In total, 39 pregnant sows (Landrace × Yorkshire; 2 parities) were randomly assigned to and maintained in the following three treatment groups (13 sows per group) over days 0 to 35 of pregnancy: (i) air cooling (AC; 26.87 ± 1.23°C), (ii) water-drip cooling (WC; 28.81 ± 0.91°C), and (iii) a lack of cooling with heat stress (HS; 30.72 ± 0.70°C). Backfat thickness was measured before and after HS. Feces were collected on day 0 and 35 d of the trial for microbiome analysis, whereas blood was taken at day 35 of pregnancy and analyzed. Reproductive performance and physiological responses were identified at day 35. Respiration rate along with rectal and skin temperatures were lower (p < 0.05) in the AC group than in the HS and WC groups. Serum blood urea nitrogen values were increased (p < 0.05) in the WC group compared with those measured in the AC and HS groups. Triiodothyronine was found at greater levels (p < 0.05) in the AC than in the HS group. Reproductive performance was not affected by the cooling systems. At the phylum level, fecal pathogenic Spirochaete and Euryarchaeota were found in higher numbers (p < 0.05) in all groups after HS. Similarly, at the genus level, the amount of Treponema was greater (p < 0.05) in all groups after HS. In conclusion, our results suggest that AC or WC can ameliorate or mitigate the adverse effects of HS on the physiological parameters of pregnant sows reared under high temperatures.

This study was conducted to provide commercial pig farms with information about group housing systems for sows in accordance with the amendment of the prohibition law for individual stalls for sows in South Korea. Therefore, this experiment was performed to compare the effects of individual stalls (IS) and group housing systems (GS) on the productivity of sows to investigate the feasibility of replacing individual stalls with group housing systems in commercial sow units. Forty primiparous sows (Landrace × Yorkshire; 210.67 ± 2.22 kg average initial body weight) were randomly assigned to four treatments with restricted feeding after 8 weeks from artificial insemination. The four treatments were (i) individual stalls (IS; housed in pen stalls), (ii) short stalls (SS; sows housed in pens with non-gated feeding stalls), (iii) free access stalls (FAS; a non-competitive housing system), and (iv) electronic sow feeders (ESF; used with radio frequency identification technology to allow individual sow management without individual confinement). All sows were transferred to farrowing crates at 110 days of gestation. There were no differences in sow productive performance, reproductive performance, and colostrum composition between IS and GS and among GS. The considered GS did not negatively affect any productivity parameters of primiparous sows compared with IS; the GS could replace IS in commercial sow units.

The present study investigated the effects of loose farrowing type during gestation and after farrowing on reproductive performance and of lactating sows. A total of 22 primiparous sows (Landrace; average initial body weights [BW], 228.54 ± 12.79 kg) were allotted to one of two treatments on the basis of body weight. Sows were divided into two experimental groups, conventional farrowing crates (CON), and loosed-farrowing pens (LFP). The experiment duration was around 38 days ranging from 10 days before parturition to 28 days after parturition. Gestating sows at the age of 105 d were placed in gestational stalls (group housing). All the sows were fed a common diet according to the National Research Council requirements for lactation. Cross-fostering was performed within 1 day of parturition. From 1 day after weaning, estrus detection was performed twice-daily (0900 and 1730 h) for 10 min by boar exposure. There were significant effects of LFP housing type on the farrowing duration, and farrowing interval. At the farrowing time, none of the litter parameters including total born, stillborn, mummy, born alive piglets and total litter weight and piglet weight were affected. There were no effects of housing type on the mortality of piglets at d 1, 3, 7, 21, and 28. In conclusion, the result of this study showed that there is no performance difference between the crated or LFP sows, which indicate that the LFP housing has the potential to be used as an alternative to the crated house without any detrimental effects in reproduction performance of lactating sows.

This study was conducted to investigate the effects of group-housing facility (GHF), compared to an individual confinement stall (CON), on the reproductive performance, behavior, and stress hormones of gestating sows. A total of 50 primiparous sows (Landrace × Yorkshire) were randomly allocated into either CON (n = 25) or GHF (n = 25) during the gestation period. One week before parturition, the sows were transferred into conventional farrowing crates, and cross-fostering was conducted within 1 d of delivery. Blood was collected for analyses of stress indices at 75 d of gestation and postpartum. Reproductive performance was estimated during the period of birth to weaning. Behavior patterns were identified at 90 d of gestation. Litter size was not different between the CON and GHF treatments. Weaning to estrus interval, however, tended to be lower in the GHF than in the CON (p < 0.1). Activity, treating, belly nosing, and exploring behaviors were observed only in the GHF group, whereas rubbing was shown only with the CON. Serum cortisol concentration was lower in the GHF than in the CON at 75 d of gestation (p < 0.05). Sows housed in the GHF showed lower epinephrine and norepinephrine concentrations than those housed in the CON at postpartum (p < 0.05). The GHF sows demonstrated more natural behavior characteristics associated with stress relief than the CON sows with no adverse effects on reproductive performance. Therefore, these results suggest that GHF could be applied as an alternative housing facility to improve animal welfare on swine farms.

This study was conducted to evaluate the effects of dietary mixture of protease and probiotics on growth performance, blood constituents, and carcass characteristics of growing-finishing pigs. A total of 48 growing pigs were randomly allotted into 2 dietary (6 pigs/pen; 4 replicates/treatment). The treatments were a diet based on corn and soybean meal (CON) and CON supplemented with 0.01% of dietary mixture of protease and probiotics (MULTI). No differences were found on growth performance (average daily gain, ADG; overall, 874.06 vs. 881.14 g/d; p > 0.05), blood constituents (white blood cell, WBC; phase I, 17.51 vs. 19.96 × 103 /µL; phase II, 19.65 vs. 21.95 × 103 /µL; p > 0.05), and carcass characteristics during overall experimental period between CON and MULTI. In conclusion, the addition of dietary mix-ture of protease and probiotics in growing-finishing pig diet did not have any beneficial effects.

This experiment was conducted to evaluate the effects of dietary protease on growth performance, blood constituents, and carcass characteristics of growing-finishing pigs. A total of 48 growing pigs (initial body weight, 34.8 ± 0.62 kg) were randomly assigned to 2 dietary treatments (6 pigs/pen; 4 replicates/treatment). The treatments were a diet based on corn and soybean meal (CON) and CON supplemented with 0.01 % of protease (PRO). Pigs were fed respective dietary treatments with a 2-phase feeding program for 12 weeks. Pigs fed PRO had higher average daily gain (ADG; phase I, 866.38 vs. 821.75 g/d; overall, 910.96 vs. 866.30 g/d; p < 0.05) and gain to feed ratio (G:F; phase I, 0.345 vs. 0.363 g/g; p < 0.05) than those fed CON. However, there were no differences on blood constituents and carcass characteristics between CON and PRO of growing-finishing pigs. In conclusion, dietary protease supplementation in the typical diet for growing-finishing pigs improved growth rate.

PURPOSE: Tubular carcinoma (TC) of the breast is an uncommon histological subtype of invasive breast cancer with an excellent prognosis compared with standard invasive ductal carcinoma. Recent studies suggested a possible precursor role for low grade ductal carcinoma in situ (DCIS) in the development of TC. The goal of this analysis was to understand the clinicopathologic features and outcomes of TC by comparing TC with DCIS. METHODS: A retrospective review identified 70 patients with TC and 1,106 patients with DCIS between 1995 and 2011. Student t-test and Fisher exact test were used to compare the clinicopathologic characteristics of TC patients with those of DCIS patients. The Kaplan-Meier method and Cox regression analysis were used to determine disease-free survival (DFS) rates. RESULTS: Compared to DCIS, TC exhibited favorable clinicopathologic characteristics such as a lower nuclear grade (92.3%), higher expression of hormonal receptors (estrogen receptor-positive, 92.9%; progesterone receptor-positive, 87.0%), and less frequent overexpression of human epidermal growth receptor 2 (12.9%). DFS did not differ significantly between the TC and DCIS groups (5-year DFS, 100% vs. 96.7%; 10-year DFS, 92.3% vs. 93.3%; p=0.324), and cancer-specific deaths were not noted in either group. However, axillary lymph node involvement was observed in six (8.6%) of the 70 patients with TC. Three of these patients had small tumors (≤1 cm). CONCLUSION: In our study cohort, TC was associated with an excellent prognosis and a low rate of lymph node metastasis. However, lymph nodes metastases were found even in patients with small tumors (≤1 cm). Axillary staging must be considered for all patients with TC of the breast.
